Abstract:Objective:To observe the effects of eye acupuncture on cognitive impairment,inflammatory factors, and microvessel density in rats with ischemic stroke (IS). Methods:A total of 60 healthy male SPF-grade SD rats were randomly divided into the control group, the sham operation group, the model group, the body acupuncture group, and the eye acupuncture group,with 12 rats in each group. Except for the control group and the sham operation group, rats in the other three groups were used to establish IS models. After modeling,the rats in the body acupuncture group received acupuncture at Baihui (GV20) point, and those in the eye acupuncture group were treated at periocular acupoint regions including the liver region, kidney region, upper energizer region, and lower energizer region. The neurological deficit scores, cognitive function, inflammatory factor levels, cerebral infarct volume, brain tissue pathological changes, and microvessel density were compared among all groups. Results: After intervention, compared with the control group and the sham operation group,the neurological deficit scores in the model group were increased (P<0.05), and compared with the model group, the neurological deficit scores in the body acupuncture group and the eye acupuncture group were decreased (P<0.05), with the eye acupuncture group presenting lower scores than the body acupuncture group (P<0.05). No significant differences were observed in escape latency, platform crossing times,levels of interleukin-6( IL-6),interleukin-1β( IL-1β),tumor necrosis factor-α( TNF-α), and the number of CD31-positive cells between the control group and the sham operation group (P>0.05). Relative to the control group and the sham operation group, the model group had prolonged escape latency, increased platform crossing times, increased levels of IL-6, IL-1β, and TNF- α, enlarged cerebral infarct volume, and reduced number of CD31-positive cells,with all differences statistically significant (P<0.05). Compared with the model group, the body acupuncture group showed shortened escape latency, decreased platform crossing times, decreased IL-6, IL-1β, and TNF-α levels and cerebral infarct volume, and increased number of CD31-positive cells (P<0.05). Furthermore,the eye acupuncture group achieved shorter escape latency,less platform crossing times,lower IL-6, IL-1β,and TNF-α levels,smaller cerebral infarct volume,and more CD31-positive cells than the body acupuncture group (P<0.05). Conclusion: Eye acupuncture can effectively improve cognitive function and relieve neurological deficits in IS rats. It exerts protective effects against ischemic brain injury by down-regulating the levels of inflammatory factors such as IL-6, IL-1β, and TNF- α to alleviate cerebral inflammatory responses, reduce cerebral infarct volume,and enhance cerebral microvessel density.
